ABSTRACT

OBJECTIVE@#To understand the genotypes and distribution characteristics of thalassemia in Baise, Guangxi Zhuang Autonomous Region, to provide references for the prevention and diagnosis of thalassemia in the region and improve the quality of eugenics.@*METHODS@#3 482 pregnant women and their spouses from January 2019 to August 2019 in Baise Maternal and Child Health Hospital for prenatal genetic diagnosis were selected, α, β- thalassemia genes were detected by Gap-PCR, PCR and DNA reverse dot hybridization, cases carrying thalassemia gene were confirmed and statistical analyzed.@*RESULTS@#2 260 samples (64.90%) carrying thalassemia gene were found, among which 1 459 cases (64.56%) were diagnosed as α- thalassemia, 617 cases (27.30%) as β- thalassemia, 184 cases (8.14%) as α complex β- thalassemia. Among 1 459 α- thalassemia genes, --SEA /αα(637 cases, 43.66%), -α3.7 /αα (306 cases, 20.97%), -αCS /αα(143 cases, 9.80%), -α4.2 /αα(124 cases, 8.50%) and -αWS /αα(77 cases, 5.27%) were the most common, while among 617 β- thalassemia genes, CD17 (229 cases, 37.12%), CD41-42 (213 cases, 34.52%), IVS-I-1 (41 cases, 6.65%), βE (38 cases, 6.16%) and CD71-72 (34 cases, 5.51%) were the most common. And --SEA /αα/ CD17 (24 cases, 13.04%), -α4.2 /αα/ CD17 (13 cases, 7.07%), -α3.7 /αα/ CD41-42 (12 cases, 6.52% ) and --SEA /αα/ CD41-42 (12 cases, 6.52%) were mainly found in 184 cases of α complex β - thalassemia.@*CONCLUSION@#Genotyes of thalassemia in Baise, Guangxi Zhuang Autonomous Region are complex and diverse. The prenatal screening and diagnosis of thalassemia in the region should be strengthened in accordance with the characteristics of genetypes in the region, in order to reduce birth defects and improve eugenics quality.

ABSTRACT

OBJECTIVE Human metapneumovirus (hMPV) is semblable to respiratory syncytial virus (RSV) which causes respiratory infections typically characterized by cough, runny nose, fever, and nasal congestion but sometimes progressing to bronchiolitis and pneumonia. Whereas, there is no corresponding drug to inhabit the virus. Studies of new compounds with potential anti-HMPV activity could produce clinical value. Chinese herbal medicine played a great role during COVID-19, therefore we choose some small molecular (JH001) extracted from botany to investigate therapeutic effect on hMPV and the underlying mechanisms. METHODS In this study, 16HBE cells were used as a model to explore in vitro antiviral effect. Cytotoxicity assays were performed before the antiviral tests, cell viability of 16HBE cells handled by different concentration of JH001 was estimated by Cell Counting Kit-8 (CCK-8). Then RT-qPCR, immunofluores?cence, and flow cytometer were used to test the viral titer after cells infected with hMPV. Eventually, 6-8 weeks mice were infected intranasally with 60 μL of hMPV, the control group was treated with 0.9% saline water, other groups were administered with JH001 and ribavirin, then the lung virus titer and protective effect in lung were judged. RESULTS The obtained JH001 exhibited no cytotoxicity to 16HBE cells during 6.25 - 200 μmol · L-1. RT-QPCR demonstrated that JH001 showed obvious inhabitation to the viral replication and showed great significance compared with saline. And fluo?rescence exhibited distinct decrease of hMPV-N protein, flow cytometer results showed that MFI decrease evidently. Sig?nificant reduction of N-gene expression was observed in those mice treated with JH001 compared with saline group, which indicated that JH001 probably had protective and therapeutic effect on viral replication. CONCLUSION This study illustrated that JH001 might be a promising option for small molecular against hMPV and JH001 might be worthy of fur?ther development and used as a potential therapeutic strategy for other respiratory viruses in the future.

ABSTRACT

Studies with electroencephalography, transcranial Doppler (TCD), magnetic resonance, positron emission tomography (PET), and single-photon emission computed tomography (SPECT) have achieved some positive results for depression in children and adolescents. Electroencephalographic studies found that the patients' α, β, δ, θ waves, and P300, P200, N100 and N200 in event-related potential (ERP) were significantly different from typical ones, and cortical activation area was uneven. TCD, magnetic resonance, PET and SPECT studies found that the patients with depression occurred cerebral blood flow abnormalities in frontal lobe and temporal lobe. Magnetic resonance studies found that there were brain structural and metabolic abnormalities in frontal lobe, left fusiform gyrus and anterior cingulate cortex, and N-acetyl aspartate, creatine, choline and glutamate might be a part of the cause of depression.

ABSTRACT

Objective: To overall evaluate the mutual detoxication mechanism of Tripterygii Radix et Rhizoma(LGT) compatible with Lysimachiae Herba(JQC) in tumor-bearing state.Method: Twelve differentially characteristic components before and after compatibility were used as chemical composition spectrum,six indicators including serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),creatinine(Cr) and urea nitrogen(BUN),malondialdehyde(MDA) levels in the liver and kidney tissues were used as attenuation spectrum,and twelve biological indicators including glutathione(GSH),glutathione-S-transferase(GST),glutathione peroxidase(GPx),superoxide dismutase(SOD),catalase(CAT) and interleukin(IL)-10 in the liver and kidney were used as the biological information spectrum.Mutual detoxication mechanisms of LGT compatible with JQC in tumor-bearing state were overall evaluated by principal component analysis(PCA),and the contribution of chemical components and biological indicators to mutual detoxication was further evaluated by gray correlation analysis(GCA) of "chemical composition spectrum-attenuation spectrum-biological information spectrum".Result: Compared with the model group,the attenuation spectrum scores Z values of S180(Z1 value) and H22(Z3 value) increased significantly after LGT being used alone(PZ1 value and Z3 value caused by LGT when the ratio of LGT and JQC was 4:1,2:1,1:1,1:2 and 1:4(PZ values(Z1 value and Z3 value) of LGT-JQC in the mass ratios including 4:1,1:1,1:2 and 1:4 was significantly higher than that in the ratio of 2:1(PZ values of the bioinformatics scores in the S180(Z2 value) and H22(Z4 value) tumor-bearing state,these two values were significantly increased after compatibility with JQC.The chemical components contributing the most to the attenuating effect of S180 and H22 in tumor-bearing state were 3# and 10#,respectively.The most important biological indicators were kidney GPx and renal GSH.Conclusion: LGT combined with JQC in the mass ratio of 4:1-1:4 can attenuate LGT-induced subacute toxicity in S180 and H22 tumor-bearing state,and the best ratio of such effect is 2:1.The attenuating effect reflects the thought of "there is no reason why there is no meteorology".The mechanism of attenuating action involves antioxidative damage and anti-inflammatory reaction of the liver and kidney,especially the renal GPx(S180) and renal GSH(H22) as the greatest contribution to the detoxication mechanism.

ABSTRACT

<p><b>Background</b>Percutaneous transluminal septal myocardial ablation (PTSMA) and modified Morrow septal myectomy (MMSM) are two invasive strategies used to relieve obstruction in patients with hypertrophic cardiomyopathy (HCM). This study aimed to determine the clinical outcome of these two strategies.</p><p><b>Methods</b>From January 2011 to January 2015, 226 patients with HCM were treated, 68 by PTSMA and 158 by MMSM. Both ultrasonic cardiograms and heart functional class were recorded before, after operations and in the follow-up. Categorical variables were compared using Chi-square or Fisher's exact tests. Quantitative variables were compared using the paired samples t-test.</p><p><b>Results</b>Interventricular septal thickness was significantly reduced in both groups (21.27 ± 4.43 mm vs. 18.72 ± 4.13 mm for PTSMA, t = 3.469, P < 0.001, and 21.83 ± 5.03 mm vs. 16.57 ± 3.95 mm for MMSM, t = 10.349, P < 0.001, respectively). The left ventricular outflow tract (LVOT) pressure gradient (PG) significantly decreased after the operations in two groups (70.30 ± 44.79 mmHg vs. 39.78 ± 22.07 mmHg for PTSMA, t = 5.041, P < 0.001, and 74.58 ± 45.52 mmHg vs. 13.95 ± 9.94 mmHg for MMSM, t = 16.357, P < 0.001, respectively). Seven patients (10.29%) in the PTSMA group required a repeat operation in the follow-up. Eight (11.76%) patients were evaluated for New York Heart Association (NYHA) III/IV in the PTSMA group, which was significantly more than the five (3.16%) in the same NYHA classes for the MMSM group at follow-up. Less than 15% of patients in the PTSMA group and none of the patients in the MMSM group complained of chest pain during follow-up.</p><p><b>Conclusions</b>Both strategies can not only relieve LVOT PG but also improve heart function in patients with HCM. However, MMSM might provide a more reliable reduction in gradients compared to PTSMA.</p>

Panax quinquefolium and Panax ginseng were investigated using non-linear chemical fingerprint technology, and a novel method to identify two ginsengs and different producing areas of P. quinquefolium was put forward. The non-linear chemical fingerprints of P. quinquefolium(collected from Canada, Jilin and Shaanxi)and P. ginseng (collected from Jilin)were obtained by reactions took place in the system of "H2SO4-MnSO4-CH3COCH3-NaBrO3" and their system similarities were evaluated. In the meantime, the quality difference in P. quinquefolium from different producing areas was evaluated using HPLC to determine the contents of main 7 ginsenosides. As a result, the non-linear chemical fingerprints exhibited a good reproducibility and characteristic when dosage of detection was 0.4 g and the reaction temperature was 37 ℃. The fingerprint characteristics of P. quinquefolium were obviously different from P. ginseng. The two species of ginsengs could be distinguished by the visual fingerprint characteristics, and P. quinquefolium from different producing areas were identified by the system similarities. Furthermore, HPLC analysis showed that the quality P. quinquefolium from different producing areas was varied, which indicated that rapid identification and quality evaluation of P. quinquefolium become very important and necessary. Compared with HPLC technology, non-linear chemical fingerprint is a more convenient, rapid and economic technology. This study provides a novel strategy to distinguish and evaluate P. quinquefolium and P. ginseng, which will provide a reference for the quality evaluation and control of Chinese medicine.

ABSTRACT

Objective: To rapidly identify the chemical constituents of Paeonia delavayi var. lutea roots by ultra-high performance liquid coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MSE) and UNIFI informatics platform. Methods: Time-dependent MSE data-acquistion mode was applied to acquire mass spectrometric data, and then the chemical constituents were identified by automatic identification and artificial identification. Results: Totally 57 compounds were identified, including monoterpene glycosides, phenolic acids, tannins, paeonols, and triterpenes. Conclusion: UPLC-Q-TOF-MSE combined with UNIFI database could be used to rapidly and comprehensively characterize the chemical constituents of P. delavayi var. lutea roots. This study provides a reference for quality control of P. delavayi var. lutea roots and clarifying the material basis of its efficacy.

ABSTRACT

A quantitative analytical method of ultra-high performance liquid chromatography (UPLC) was developed for simultaneously determining twelve components in Tibetan medicine Zuozhu Daxi. SIMPCA 12.0 software was used a principal component analysis PCA) and partial small squares analysis (PLSD-DA) on the twelve components in 10 batches from four pharmaceutical factories. Acquity UPLC BEH C15 column (2.1 mm x 100 mm, 1.7 µm) was adopted at the column temperature of 35 °C and eluted with acetonitrile (A) -0.05% phosphate acid solution (B) as the mobile phase with a flow rate of 0. 3 mL · min(-1). The injection volume was 1 µL. The detection wavelengths were set at 210 nm for alantolactone, isoalantolactone and oleanolic; 260 nm for trychnine and brucine; 288 nm for protopine; 306 nm for protopine, resveratrol and piperine; 370 nm for quercetin and isorhamnetin. The results showed a good separation among index components, with a good linearity relationship (R2 = 0.999 6) within the selected concentration range. The average sample recovery rates ranged between 99.44%-101.8%, with RSD between 0.37%-1.7%, indicating the method is rapid and accurate with a good repeatability and stability. The PCA and PLSD-DA analysis on the sample determination results revealed a great difference among samples from different pharmaceutical factories. The twelve components included in this study contributed significantly to the quantitative determination of intrinsic quality of Zuozhu Daxi. The UPLC established for to the quantitative determination of the twelve components can provide scientific basis for the comprehensive quality evaluation of Zuozhu Daxi.

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect of Fuzheng Sanjie recipe in regulating tumor-associated macrophages (TAMs) in Lewis lung cancer mice.</p><p><b>METHOD</b>Efforts were made to establish the Lewis lung cancer mouse model, weigh tumors and calculate the anti-tumor rate. The immunohistochemical method was used to examine the infiltration degree of CD68 + in tumor tissues in each group. ELISA was used to examine the content of IFN-γ, TGF-β, IL-4, IL-13, IL-6, IL-10, IL-12, TNF-α in mice serum.</p><p><b>RESULT</b>Compared with the tumor-bearing model group, all of the other groups showed higher tumor inhibition rates, i. e. 50.28% for the DDP group, 34.37% for the TCM-preventing group and 66.76% for the Chinese and western medicine group, with statistical difference (P < 0.05), but without statistical difference in the infiltration degree of CD68+. The expressions of the IFN-γ, IL-6, IL-12 in tumor-bearing groups were lower than that in the blank control group, but with higher contents of IL-4, IL-13, TGF-β. Intervened with different drugs, there were significant differences in content among some relevant cytokines (P < 0.05), as well as statistical differences among the TCM prevention group, the Chinese and western medicine group and the tumor-bearing control group (P <0. 05) , but without statistical difference in TNF-α and IL-10 content from the tumor-bearing control group (P < 0.05).</p><p><b>CONCLUSION</b>Fuzheng Sanjie recipe could reverse the immune remodeling effect and control the tumor growth by down-regulating the expressions of IL-4, IL-13, TGF-α in lung cancer immune microenvironment and up-regulating the expression of IFN-γ.</p>

ABSTRACT

Objective To explore the relationship between substance P(SP),somatostatin(SS) expression and change of morphology structure in jejunum of arsenism rats.Methods Acoording to sex and body mass,forty five clean grade SD rats were divided into control(0.0 mg.kg-1.d-1),low-dose arsenic(0.4 mg.kg-1.d-1) and high-dose arsenic(10.0 mg.kg-1.d-1) groups,n =15.The rats in low-and high-dose groups were treated with As2O3(2,50 mg/L) through drinking water for 4 months,respectively.Morphology changes of jejunum were observed by histological technique-HE staining and SABC immunohistochemistry.SP and SS positive cells in the jejunum were observed and counted,and its average gray value was analyzed with image analysis software (Biomias).Results Some jejunal villi were irregular in arsenism rats; with some brush border loss and irregular; goblet cells increased; infiltration of inflammatory cells in the lamina propria; and vacuoles in some intestinal gland cells.The differences of SP and SS positive cells between groups were statistically significant (F =608.54,227.59,all P <0.05).Compared with the control group (0.94 + 0.21,1.14 + 0.14),SP and SS positive cells in low-and highdose arsenic groups(1.85 + 0.25,1.83 + 0.24 and 4.24 + 0.33,3.31 ± 0.41) were significantly higher(all P <0.05),and high-dose arsenic group was significantly higher than the low-dose arsenic group(all P < 0.05).The differences of average gray values of SP and SS positive cells between groups were statistically significant(F =68.43,26.57,all P < 0.05).Compared with the control group(133.76 ± 3.61,137.57 ± 5.49),SP and SS positive cells in low-and high-dose arsenic groups(125.13 + 2.35,131.28 ± 5.66 and 118.30 ± 4.58,124.03 ± 3.94) were significantly lower(all P < 0.05),and high-dose arsenic group was significantly lower than the low-dose arsenic group (all P < 0.05).Conclusions Up-regulation of SP,SS may be related to jejunal mucosal injury and morphology structure in arsenic poisoning rats.

ABSTRACT

ObjectiveTo investigate the expression and relevant function of protein kinase C (PKC)-α in kidney of rat with chronic arsenic poisoning.MethodsTotally 60 healthy SD rats of clean grade were randomly divided by body weight into 3 groups:high-dose arsenic exposure group (10.0 mg/kg),low-dose arsenic exposure group (0.4 mg/kg),and control group.The rats were exposed by drinking arsenic solution which was mixed with distilled water.Rats were weighed every 10 days and dose volume of arsenic solution was adjusted.After continuous exposure for 4 months,blood and urinary arsenic were determined.Rat kidneys were taken and stained by Immunohistochemistry SABC.PKC-o positive cells in the kidney were observed and counted,and its average gray value was analyzed with image analysis software (Biomias).ResultsProximal tubules PKC-α-positive cell count [(3.62 ± 1.90),(10.07 ± 3.22)/field],glomerular PKC-α-positive cell count [(3.62 ± 1.90),(10.07 ± 3.22)/field]in high and low arsenic group of SD rat kidney were lower than those of the control group [(60.00 ± 9.63),(18.57 ± 2.71/field,all P < 0.05]; both urinary arsenic level[(7366.62 ± 1086.50),(1744.31 ± 300.12)μg,/L]and blood arsenic level [(31.59 ± 9.24),(16.58 ± 2.08)μg/L] in high-dose and low-dose groups were higher than those of the control group [(18.97 ± 3.58),(18.97 ± 3.58)μg/L,all P < 0.05] ; the average gray values of SD rat kidney proximal tubule,glomerular PKC-o positive cells in high-dose and low-dose groups( 142.79 ± 11.16,122.15 ±5.91 ) were higher than that of the control group (114.33 ± 6.70,all P < 0.05).ConclusionsArsenic can decrease SD rat kidney PKC-α -positive cells.The regulatory function of PKC-o in inhibiting cell apoptosis of kidney of rats with arsenic poisoning is weakened.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the color changes of nickel-titanium (Ni-Ti) orthodontic wires induced by fluoride.</p><p><b>METHODS</b>Each 40 samples of 4 kinds of nickel-titanium orthodontic wires (IMD, SL, TP, YY) were put into artificial saliva with sodium fluoride at different F concentrations (0.05%, 0.10%, 0.15%) and artificial saliva, separately. Dislodged the species and immersed them into artificial saliva with sodium fluoride for 3 minutes, then washed them with deionized water, and put back to artificial saliva, this course should be repeated 3 times per day. The whole immersion test lasted for 28 days. The color and micro-morphous changing were observed.</p><p><b>RESULTS</b>The color changes came to be more serious as the F- concentration growing (P < 0.05). At the same F- concentration, the color changes of IMD and YY were higher than SL and YP (P < 0.01), while there were no significant difference between IMD and YY, SL and TP (P > 0.05).</p><p><b>CONCLUSION</b>Fluoride can change the color of Ni-Ti wire. Different Ni-Ti wires get different changes at the same F- concentration.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the microscopic characteristic of the dentinal surface and cross section after using dental desensitizer, and evaluate the effect of the dental desensitizer on the shear strength of resin cements.</p><p><b>METHODS</b>To use the dental desensitizer (Gluma, Single Bond 2, BisBlock) individually on the prepared human bicuspid teeth which were extracted for orthopaedics. The microscopic characteristic of the dentinal surface and cross section were observed with scanning electron microscope (SEM). The adhered posts to the teeth pretreated with desensitizer were bonded with resin cements. The shear strength was measured and data was analyzed by SPSS 13.0 statistics software.</p><p><b>RESULTS</b>The orifices of dentinal tubule were mostly occluded in Gluma samples and partly occluded in BisBlock samples. Resin layer was observed in Single Bond 2 samples. BisBlock group gathered the best shear strength, the shear strength value was (13.04+/-2.76) MPa. Marked statistic difference were found when compared with control group.</p><p><b>CONCLUSION</b>Gluma, Single Bond 2 and BisBlock can block the orifices of dentinal tubule. BisBlock enhanced the bonding strength of the resin cements.</p>

ABSTRACT

<p><b>AIM</b>To investigate the roles of alpha1 and alpha2 receptors in the nucleus tractus solitarius (NTS) in the carotid baroreflex (CBR) resetting induced by the intracerebroventricular injection (ICV) of histamine (HA).</p><p><b>METHODS</b>The left and right carotid sinus regions were isolated from the systemic circulation in 25 Sprague-Dawley rats anesthetized with pentobarbital sodium. The intracarotid sinus pressure (ISP) was altered in a stepwise manner. ISP-mean arterial pressure (MAP) relationship curve and its characteristic parameters were constructed by fitting to the logistic function with five parameters. The changes in CBR performance induced by ICV HA and the effects of pretreatment with alpha1 or alpha2 receptor antagonist into the NTS on the responses of CBR to HA were examined.</p><p><b>RESULTS</b>ICV HA (60 micromol x L(-1) in 5 microl) significantly shifted the ISP-MAP relationship curve upwards (P < 0.05) and moved the middle part of ISP-Gain relationship curve downwards (P < 0.05), and reduced the MAP range and maximum gain (P < 0.05). The pretreatment with phenoxybenzamine (PBZ, a selective antagonist of alpha1 receptor, 3 micromol x L(-1) in 500 nl) or yohimbine (YOH, a selective antagonist of alpha2 receptor, 2.5 micromol x L(-1) in 500 nl) into the NTS could obviously intensify the above-mentioned changes in CBR performance induced by HA, but the intensive effect of PBZ was less remarkable than that of YOH (P < 0.05).</p><p><b>CONCLUSION</b>The intracerebroventricular administration of HA results in a rapid resetting of CBR and a decrease in reflex sensitivity, and the functions of alpha1 and alpha2 receptors in the NTS might weaken CBR resetting induced by ICV HA. Furthermore, alpha2 receptor in the NTS might play an more important role in modulating the responses of CHR to HA.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the pathogen of opium poppy downy mildew and its biological characteristic for further research on the disease.</p><p><b>METHOD</b>Development of the disease was observed systematically in the field. Germination rate of sporangium in different temperature, pH and nutrition was examined with suspending-drop method. Slide-germination method was used to observe its germination in different humidity maintained by different concentration of H2SO4.</p><p><b>RESULT AND CONCLUSION</b>The disease manifests itself in two forms: severely infected plants (systematic infection) and leaf spots (nonsystematic infection). Sporangia of the pathogen are oval or globular, thin walled, smooth, hyaline, with 7.74-16.34 microns diameter in base 1 and 8.34-15.05 microns in base 2.0 ospores are light yellow with 33.87-70.54 microns x 19.34-62.64 microns in base 1 and 36.85-49.68 microns x 42.08-55.76 microns in base 2. Conidiophores are stout, erect, whose branching times and length are different between those in base 1 and those in base 2. Sporangia sprot directly in two hours. Film of water is necessary for sporangium to sprot. The optimum temperature range of sporangium sprot is 12-21 degrees C, the best being 16 degrees C, the pH range is 4.53-9.18 the best optimum at pH 7.38, and the extract of leaf of 1:5 is good for its germination.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To screen effectual fungicides for field control because of the seriousness of opium poppy mildew and importance of chemical control on plant diseases.</p><p><b>METHOD</b>Seven fungicides were screened in Lab experiment and field test during 1996-1997.</p><p><b>RESULT AND CONCLUSION</b>All of them and their different dosages were effective to control conidia of Peronospora arborescens. Among them, 72.2% propamocarb of 1203 and 902.5 ppm were the most effective both in Lab experiment and field test with efficacy 79.91% and 79.33% respectively in field test, and the efficacy of other fungicides was over 50%. Seven fungicides tested can be used to control nonsystematic symptom of opium poppy mildew.</p>
